Voraussetzungen: Totipotenz/Omnipotenz von Pflanzenzellen
Totipotenz:
Fähigkeit von einer Zelle sich zu jeder beliebigen Zelle, Gewebe oder sogar zu einem
ganzen Organsmus zu regenerieren
– Menschen und Tiere => nur embrionale Zellen sind maximal totipotent bis zu
einer Zellteilungsphase von 4-8-Zellen
– Pflanzen => alle Pflanzenzellen sind totipotent?

Kallus: nicht differenzierter Pflanzen-Zell-Komplex (parenchymatisch) mit nicht
gerichtetem Wachstum. Die Bildung von Kallus wird verursacht durch:
– Verwundung (Schnitt, Verwundung durch Insekten oder Fraß ,
Windbruch, …)
=> Kallus verschließt die Wunden (Narben-Gewebe)
– Holzpflanzen (lignifizierte) Pflanzen: Kallus hautsächlich aus Kambium
– Krautige Pflanzen: Kallus hauptsächlich aus dem Parenchym

• Es gibt in der Züchtung viele Probleme:
• Bei Genverschmelzungen (=> Offsprings from a cross)
=> hohe genetische Variabilität
• (Mono-)Haploide Pflanzen besitzen nur ein Basis-Genome (x)
Vorteile von haploiden Pflanzen in der Züchtung?
• rezessive Mutationen werden sichtbar
• Kleinere Pflanzen (bei Zierpflanzen erwünscht)
• Längere Blüte (bei Zierpflanzen erwünscht)
• Kürzere Produktionszeit bis zur Reife
• Direkte Entwicklung von homozygoten Individuen
• Basis-Material für doppelt-haploide Pflanzen (Homodiploide)
Aber: in haploiden Pflanzen:
• Meiose ist sehr ungleichmäßig
• Bei der Metaphase I ist der Spindelapparat sehr unorganisiert. Bei der Anaphase I ist die
Verteilung der Chromosomen zu den entgegengesetzten Polen teilweise ungeordnet
• Nachkommen von Doppel-Haploiden Pflanzen sind dagegen bei allen wichtigen Merkmalen
genetisch gleichmäßiger.

polar migration of chromosomes (colchicin ligates to microtubuli subunits)

2. No separation of doughter-chromatids caused by missing spindle fibers (Anaphase)
3. At cell division => 1 cell with nucleus 1 cell without nucleus (nonviable)
4. First cell doubeling of chromatides (Interphase) => polymerisation
problems:
• Not all organs show doubling in the same degree
• Zea maize => doubling chromosome number in the tassel or the ear, but often not in both,
which will make self-pollination impossible (Wan et al., 1989)
• high mortality
• abnormal plant development
Alternatives:
• doubling before plantlet regeneration
• colchicine treatment of embryogenic, microspore-derived haploid calli => stable doubled-
haploid maize plants at high frequency (Wan et al. (1989) and Wan and Widholm (1995))
• colchicine together with a 7-days cold shock to cultured maize anthers => increase in
chromosome doubling in microsporederived plants (Saisingtong et al., 1996; Antoine-
Michard and Beckert, 1997).
