• Spectrin (peripheral protein on cytosolic surface) • Ankyrin attaches spectrin (& the cytoskeleton) to
the inner surface to the red cell plasma membrane
• Glycophorin also attaches the cytoskeleton to the red cell plasma membrane

Ankyrin binds to spectrin and to the cytosolic domain of band 3 protein, a transmembrane protein
Spectrin also attached to band 4.1 protein, which is in turn anchored to the membrane by association with the cytosolic domain of glycophorin, a transmembrane protein

Function of Band 3 Protein
• Catalyzes the coupled transport of anions – Red blood cells carry oxygen from the lungs to tissues
• Also carry CO2 from tissues to the lungs (herein is the role of Band 3 protein)
– CO2 only sparingly soluble in water, hence carried as bicarbonate (HCO3
-)
H20 + CO2 ↔ HCO3- + H+
– Band 3 protein transports HCO3- across the membrane in
exchange for Cl- • By making the cell membrane permeable to HCO3
-, Band 3 protein increases the amount of CO2 the red blood cell can carry.
